چکیده

The important role of amino acids in spore germination was established by Hills (1949) and Stewart and Halvorson (1953). Their studies, however, were limited to the aerobic forms because the anaerobes could not be made to produce sufficient quantities of spores in media within a specified time. Using an autoclaved 1 per cent solution of glucose phosphate, Wynne et al. (1954) germinated anaerobic spores in 1 hr, but rapid germination in a medium of well-defined nutrients has not been reported. The investigation reported here had three phases: (1) the development of media and methods for growing and harvesting anaerobic spores, (2) the determination of the optimum conditions and minimal nutritional requirements for their rapid germination, and (3) a study of the effect of inhibitors on germination.

Effect of sodium nitrite, sodium chloride , and sodium nitrate on germination and outgrowth of anaerobic spores.

The effects of meat-curing agents on germination and outgrowth of putrefactive anaerobe 3679h (PA 3679h) spores were studied in microcultures. Nitrite concentrations up to 0.06% at pH 6.0 or between 0.8 and 1% at pH 7.0 allowed emergence and elongation of vegetative cells but blocked cell division. The newly emerged cells then lysed. With more than 0.06% nitrite at pH 6.0 or more than 0.8 to 1%...

Triggering Germination Represents a Novel Strategy to Enhance Killing of Clostridium difficile Spores

BACKGROUND Clostridium difficile is an anaerobic, spore-forming bacterium that is the most common cause of healthcare-associated diarrhea in developed countries. Control of C. difficile is challenging because the spores are resistant to killing by alcohol-based hand hygiene products, antimicrobial soaps, and most disinfectants. Although initiation of germination has been shown to increase susce...

The nutritional requirements for germination and outgrowth of spores and vegetative cell growth of some aerobic spore forming bacteria.

During the last 10 years the subject of bacterial spore germination has been intensively investigated. In most of these studies it has been found that bacterial spores are capable of germination under conditions which do not permit the germinated form to survive (Schmidt, 1955; Stedman, 1956). Hills (1949) pointed out that a distinction must be drawn between the requirements for germination and...

Sequence of events during germination of putrefactive anaerobe 3679 spores.

The sequence of changes during germination of putrefactive anaerobe 3679h spores was studied under aerobic conditions in a solution containing l-alanine and sodium pyrophosphate. Evidence that specific changes occurred in two distinct regions of the spore is given by data on several criteria that were used to measure germination. During the initial stage of germination, the absorbancy decreased...
